The Development of Animal Models for Diabetes and Hypertension I. Blood pressure changes in streptozotocin-induced insulin-dependent and non-insulin-dependent diabetic SHR rats



Abstract
Hypertension is frequently accompanied in diabetic patients. Also hypertension is known as one of risk factors for the progression of diabetic vascular disease. Even though there has been a few animal models for diabetes and hypertension, the changes of blood pressure is not still clear in diabetic animal models. We examined the changes of blood pressure in STZ-induced IDDM and NIDDM in SHR rats to establish animal models for diabetes and hypertension. Insulin dependent diabetes mellitus was induced by intaperitoneal injection of streptozotocin(60§·/Kg) at 8 weeks of age in spontaneously hypertensive rats(SHR). Only vehicle were injected to control groups. Non-insulin dependent diabetes mellitus was induced by intaperitoneal injection of streptozotocin (60§·/Kg) at 2day old age in SHR and its normotensive strain, WKY rats. We examined the changes of systolic and diastolic blood pressure, heart rates and blood glucose levels bimonthly until 10 month-old age. Body weights of STZ-SHR were decreased and blood glucose levels were rapidly increased to 500§·/§£. The systolic and diastolic blood pressure, heart rates of STZ-SHR were decreased than those of SHR rats. By contrast, systolic and diastolic blood pressure of SHR were increased with aging. In NSTZ-SHR, systolic and diastolic blood pressure, heart rates of STZ-SHR were increased with aging. There was not significant differences between SHR and NSTZ-SHR rats.
STZ-SHR rats is not useful animal model for diabetes and hypertension. But NSTZ-SHR rats could be used as an animal model for diabetes and hypertension. The resaon for differences of changes of blood pressure between STZ-SHR and NSTZ-SHR should be elucidated in further studies.
